PTMScan® Pathway Based Enrichment Kits

PTMScan Pathway Enrichment Bead

Bridging Discovery and Targeted Approaches

Perform mass spectroscopy-based screening and quantification of defined sets of thousands of protein-sites in important signaling pathways with PTMScan® pathway based enrichment kits. These kits include the same antibodies and technology used in the PTMScan® Direct Services, employing multiplexed beads labeled with standard site-specific antibodies for defined targets within known pathways of interest.

Coverage includes both total and phosphorylated protein sites at key signaling nodes critical for drug discovery, biomarker discovery, and diagnostic development. This approach offers comprehensive analysis of signaling with reduced likelihood of missing important data points, with the flexibility of customizable, targeted follow-up experiments.

Hierarchical Clustering of Results

PTMScan Multi-Pathway Clustering

Hierarchical clustering of enriched peptides by relative abundance using the PTMScan® Multi-Pathway Enrichment Kit. Left: each row represents the results of a unique peptide against the tissue type. Right: detailed view of Akt peptides identified and quantified. Data was calculated on a 0 (white, lowest abundance) to 1 (blue, highest abundance) scale.

Bridge The Gap

The PTMScan® Multi-Pathway Enrichment kit offers the flexibility to assay signaling pathway activities across cell lines or experimental treatments with customizable follow-up experiments. For example, a small number of samples may be run in order to narrow down to the most abundant or relevant targets. The same kit can be subsequently used with a larger number of samples, focusing on the priority targets established in the first step. Validation of PTMScan® results can be performed using AQUA synthetic peptides or other peptide standards.

All PTMScan® kits come with reagents for 10 assays and detailed protocols. Purchase of the kit also includes a Limited Use License to perform PTMScan® Technology.
